Cluster Seminar by Debattama Sen (Harvard)

Speaker
Dr. Debattama Sen
Haining Lab, Dana-Farber Cancer Institute, Harvard

Title
Epigenetic Determinants of CD8+ T cell Exhaustion

Date and Time
Tuesda, June 18, 2019 at 11:00 a.m

Location
Lecture hall (basement floor)
Biomedizinisches Zentrum (BMZ), Venusberg

Host
Prof. Christian Kurts & Dr. Zeinab Abdullah

T cell exhaustion describes an acquired dysfunction common in settings of chronic viral infection. Despite clinical efforts to rescue exhaustion, the fundamental mechanisms specifying this state, and the potential for reprogramming exhausted T cells, remain poorly understood. Since the epigenome can govern cell state, we profiled accessible chromatin in exhausted CD8+ T cells. Exhausted cells acquired a state-specific landscape of enhancers that profoundly differed from functional memory. Comparison of mouse cells to those isolated from HCV and HIV infection identified a conserved epigenetic program of exhaustion. Critically, CD8+ tumor infiltrating lymphocytes (TILs) also shared epigenetic and transcriptional features with chronic viral infection, suggesting that T cell exhaustion is a fundamental adaptation to settings of chronic stimulation.

Title
Integrative Analysis of RNA Binding Protein Targets and Funtion

Date and Time
Monday, June 17, 2019 at 11:30 a.m (11:30 Hrs)

Location
Lecture hall (basement floor)
Biomedizinisches Zentrum (BMZ), Venusberg

Host
Prof. Dr. Katrin Paeschke

Dr. Markus Hafner an Earl Stadtman Tenure-Track Investigator at the National Institutes of Health in Bethesda, MD. His RNA Molecular Biology Group (RMBG) studies the impact of RNA binding proteins (RBPs) on posttranscriptional gene regulation (PTGR). PTGR summarizes various processes acting upon coding and non-coding RNA and includes RNA maturation, ribonucleoprotein (RNP) assembly, transport, translation, and turnover. The recent introduction of large-scale quantitative methods, such as next-generation sequencing (NGS) and modern mass spectrometry (MS), allows for the first time determination of the functional impact of RBPs on a systems-wide level and has sparked a renewed interest in the systematic characterization of PTGR processes. Members of the RMBG focus on elucidating the function and molecular mechanisms of RNA binding proteins involved in RNA transport, RNA stability and turnover, and RNA translation.

The Core Facility Nanobodies cordially invites you to the first event of the lecture series “Emerging Technologies” organized by the core facilities of the University of Bonn.
Christian Stigloher is an outstanding expert on advanced electron microscopy techniques such as high pressure freezing, electron tomography, and correlated light and electron microscopy (CLEM). He uses these techniques in different model systems, such as zebrafish and C. elegans, to study synaptic architecture at the nanoscale in 3D. His special research interest is the use of CLEM to combine ultrastructure analysis with the localization of molecular factors to characterize synaptic function.

The talk will be followed by snacks and drinks and the opportunity to discuss with the speaker. For more information please visit the Core Facilities website: https://cores.ukb.uni-bonn.de/events/.
